To a solution of 2-bromo-1-(4-chlorophenylmethyl)-1H-benzimidazole (0.029 g) in n-butanol (0.5 ml) was added 1-[2-[4-(2-methoxycarbonyl-2-methylpropyl)phenyl]ethyl]-1,5-diazacyclooctane (0.102 g), and the mixture was refluxed for 3 hours. The reaction mixture was quenched with a saturated aqueous NaHCO3 solution (30 ml), and the mixture was extracted with methylene chloride (20 ml). The extract was washed with water, and dried over anhydrous MgSO4. The solvent was removed under reduced pressure, and the residue was purified by medium pressure liquid column chromatography on a silica gel column using a mixture of methylene chloride, diethyl ether and methanol (1:1:0.2) as eluent to give 1-[1-(4-chlorophenylmethyl)-1H-benzimidazol-2-yl]-5-[2-[4-(2-methoxycarbonyl-2-methylpropyl)phenyl]ethyl]-1,5-diazacyclooctane (0.030 g) as a colorless oily liquid.
