Reaction Steps

A mixture of N-(3-methyl-5-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)phenyl)pyrimidin-2-amine (220 mg, 0.707 mmol), methyl 3-(4-bromo-1H-pyrazol-1-yl)cyclobutanecarboxylate (Isomer 1) (183 mg, 0.707 mmol), Pd2(dba)3 (32 mg, 0.035 mmol), X-phos (34 mg, 0.071 mmol), and cesium carbonate (691 mg, 2.12 mmol) in 1,4-dioxane (3 mL) and water (0.3 mL) was degassed via a subsurface argon sparge for 5 minutes. The mixture was then heated to 90° C. under an argon atmosphere for 2 hours. The reaction mixture was cooled to ambient temperature and diluted with ethyl acetate (100 mL) and water (25 mL). The organic layer was separated and washed with water (25 mL) and then brine (25 mL). The organic layer was dried over anhydrous magnesium sulfate, filtered, and concentrated under reduced pressure to afford the crude product residue. The crude product residue was purified by column chromatography on silica (0-100% ethyl acetate/hexanes) to afford methyl 3-(4-(3-methyl-5-(pyrimidin-2-ylamino)phenyl)-1H-pyrazol-1-yl)cyclobutanecarboxylate (Isomer 1) MS ESI calc'd. for C20H22N5O2 [M+H]+ 364. found 364. 1H NMR (500 MHz, DMSO-d6) δ 9.48 (s, 1H), 8.46 (d, J=4.5 Hz, 2H), 8.16 (s, 1H), 7.80 (s, 1H), 7.72 (s, 1H), 7.40 (s, 1H), 6.98 (s, 1H), 6.81 (t, J=5.0 Hz, 1H), 5.05-5.00 (m, 1H), 3.67 (s, 3H), 3.24-3.17 (m, 1H), 2.81-2.74 (m, 2H), 2.67-2.60 (m, 2H), 2.27 (s, 3H).
