Procedure
A phospho-vanillin reagent was prepared by dissolving 6 grams vanillin in water in a 1 liter flask and diluting to volume with water. 350 ml. of this vanillin reagent was admixed with 50 ml. of water in a 2 liter flask. Thereafter, 600 ml. of concentrated phosphoric acid, specific gravity 1.7, was added to the vanillin reagent with constant stirring. The mole ratio of vanillin to phosphoric acid in the reagent is 1.56 × 10-3.
