Reaction Steps
To a suspension of sodium hydride (8.4 mg of a 60% dispersion, 0.210 mmol) in DMF (4 mL) cooled in an ice-bath, was added ethyl [(1S)-5-(3-{[4-(1,3-benzodioxol-5-yl)-2-pyrimidinyl]amino}propoxy)-2,3-dihydro-1H-inden-1-yl]acetate (Example 291, 50 mg, 0.105 mmol). The mixture was stirred for 15 min after which iodomethane (61 mg, 0.421 mmol) was added dropwise. The mixture was then stirred at rt for 16 h. A solution of ammonium chloride (10% in water) was added to the reaction mixture after which the mixture was concentrated under reduced pressure and brought to basic pH using a saturated solution of NaHCO3. The mixture was extracted with EtOAc (2×). The combined organic layers were washed with water and brine, dried, filtered, and then concentrated under reduced pressure. The residue was purified by silica gel flash column chromatography (1:2 EtOAc/hexanes) to give the product (136 mg, 69%) as a yellow oil. 1H NMR (400 MHz, CDCl3): δ 1.33 (t, 3H), 1.70-1.79 (m, 1H), 2.15-2.20 (m, 2H), 2.33-2.51 (m, 2H), 2.70 (dd, 1H), 2.82-2.95 (m, 2H), 3.30 (s, 3H), 3.53-3.64 (m, 1H), 3.71 (q, 2H), 4.13 (t, 2H), 4.22 (q, 2H), 6.01 (s, 2H), 6.65 (d, 1H), 6.75 (s, 1H), 6.90 (d, 2H), 7.05 (d, 1H), 7.62 (d, 1H), 7.15 (s, 1H), 8.30 (d, 1H); LC-MS: RT=3.01 min, (M+H)+ 490.2.
