Reaction Steps

To a solution of 5-(2,3-dihydropyran-2-yl)-3-(N-methylpyrrolidin-3-yl)-1-benzoylindole (Example 18a, 0.158 g, 0.409 mmol) in methanol (5 mL) was added 2N NaOH (6.8 mmol) and the mixture was heated at reflux for 2 hours. The solution was cooled to room temperature and the solvent was evaporated and the product was extracted with CH2Cl2 (3×). The organic layer was dried and concentrated by reduced pressure and purified by column chromatography [10% NH3 (2M in MeOH) in CH2Cl2 ] to yield the title compound (0.035 g, 30%) as a yellow foam; HRMS-FAB+ for C18H22N2O; calc. MH+ :283.1810; found MH+ :283.1839.
