2',3'-di-O-acetyl-5'-deoxy-N4 -[(3-methoxy-2,2- dimethylpropoxy)carbonyl]-5-[2-(trimethylsilyl)ethynyl]-cytidine (100 mg, 0.18 mmol) was dissolved in methanol (4 ml), followed by addition of K2CO3 (38 mg,0.27 mmol). After stirring for 30 min at room temperature, the reaction mixture was evaporated under reduced pressure. Purification of the residue by preparative thin layer chlomatography on silica gel (using dichloromethane:methanol=20:1 as a developing solvent) gave 5'-deoxy-5- ethynyl-N4 -[(3-methoxy-2,2-dimethylpropoxy)carbonyl]cytidine (43 mg, 60%).
