Reaction Steps

The mixture of 4-chloro-5,7-dihydrothieno[3,4-d]pyrimidine (2.0 g, 11.6 mmol) and 1-Boc-piperazine (5.0 g, 26.8 mmol) in isopropanol (50 mL) was refluxed for 12 hours. The solvent was removed and the residue was subject to silica gel chromatography, eluting with hexane/ethyl acetate (1:1) to give tert-butyl 4-(5,7-dihydrothieno[3,4-d]pyrimidin-4-yl)piperazine-1-carboxylate (3.3 g, 89%). 1H NMR (CDCl3, 400 MHz) δ 8.49 (s, 1H), 4.68 (m, 1H), 4.25 (s, 2H), 4.16 (s, 2H), 1.49 (s, 9H). MS (APCI+) [M+H]+ 323.
