A glass microwave reaction vessel was charged with N-isopropyl-5-(3-(2-(isopropylamino)-6-((4-methoxybenzyl)oxy)pyrimidin-4-yl)-1-tosyl-1H-indol-5-yl)-1,3,4-oxadiazol-2-amine (100 mg, 0.150 mmol) and NaOH (0.3 mL, 3.00 mmol) in p-dioxane (1.0 mL). The reaction mixture was stirred and heated in a Initiator microwave reactor (Personal Chemistry, Biotage AB, Inc., Upssala, Sweden) at 100° C. for 1 h. The mixture was diluted with CHCl3/iPrOH (4:1) and washed with water. The organic layer was dried, filtered and concentrated to give the crude N-isopropyl-5-(3-(2-(isopropylamino)-6-((4-methoxybenzyl)oxy)pyrimidin-4-yl)-1H-indol-5-yl)-1,3,4-oxadiazol-2-amine (70 mg, 0.136 mmol, 91% yield), which was used in the next step without further purification. MS (ESI, pos. ion) m/z: 514.1 (M+1).
