5-Chloro-N-((1-(4-(2-oxopiperazin-1-yl)phenyl)-1H-1,2,3-triazol-4-yl)methyl)thiophene-2-carboxamide (23 mg, 0.055 mmol, prepared as shown in Example 60) was dissolved in 2 mL water and 0.5 mL DMSO. To it was added KOCN (23 mg, 0.28 mmol). The mixture was stirred at RT over the weekend. The title compound was the major product in this reaction and was isolated using direct reverse phase prep HPLC. MS found for C19H18ClN7O3S (M+H)+ 460.1, 462.1 (Cl pattern).
