A mixture of N-[(3-bromo-4-fluorophenyl)methyl]-N′-{[1,6-diethyl-4-(tetrahydro-2H-pyran-4-ylamino)-1H-pyrazolo[3,4-b]pyridin-5-yl]methyl}butanediamide (1.08 g, 1.832 mmol), (3-formylphenyl)boronic acid (0.330 g, 2.2 mmol), tetrakis (triphenylphosphine) palladium (0) (0.092 g, 0.08 mmol), sodium carbonate (0.530 g, 5.0 mmol) and 1,4-dioxane (12 mL) and water (4 mL) was microwaved at 140° C. for 30 min. The reaction mixture was diluted with water and extracted with EtOAc twice. The organic layer was filtered through a glass-fiber filter, washed with saturated NaCl, and then dried over Na2SO4. The organic layer was evaporated to dryness and taken up in a small amount of DCM and introduced onto a Chromatotron Plate for purification eluting with DCM-MeOH (2-7%). The product fractions were combined to give N-{[1,6-diethyl-4-(tetrahydro-2H-pyran-4-ylamino)-1H-pyrazolo[3,4-b]pyridin-5-yl]methyl}-N′-[(6-fluoro-3′-formyl-3-biphenylyl)methyl]butanediamide (910 mg, 81%) as a glassy solid. LC-MS m/z 615.4 (M+H)+, 0.82 min (ret time).
