To 5-(aminomethyl)-1,6-diethyl-N-(tetrahydro-2H-pyran-4-yl)-1H-pyrazolo[3,4-b]pyridin-4-amine (0.34 g, 1 mmol) in DCM (5.00 mL) was added 5-(methyloxy)-5-oxopentanoic acid (146.14 g, 1 mmol), HBTU (417.16 g, 1.1 mmol), and TEA (202.38 g, 2 mmol). This was stirred at room temperature for 17 h. The reaction was quenched with H2O (1 mL). The organic layer was loaded onto a Redisep silica gel cartridge (12 g) and purified by CombiFlash Chromatograph to afford the title compound (0.4499 g, 104.26%). LC-MS m/z 432 (M+H)+.
