Process (A) To a mixture of 1,1-dimethylethyl (2S)-4-{[3′-({[5-({[1,6-diethyl-4-(tetrahydro-2H-pyran-4-ylamino)-1H-pyrazolo[3,4-b]pyridin-5-yl]methyl}amino)-5-oxopentanoyl]amino}methyl)-3-biphenylyl]methyl}-2-methyl-1-piperazinecarboxylate in DCM (2 mL) was added TFA (1 mL). It was stirred at room temperature for about 1 hour. The solution was filtered. Initial purification via Gilson using TFA afforded a mixture, which was re-purified by Gilson using a NH4OH buffer to provide the desired product (20 mg). LC-MS m/z 713 (M+H)+.
