Reaction Steps
Into a 10-mL round-bottom flask equipped with a magnetic stirbar was added sodium chlorite (28 mg, 0.31 mmol), 2-methyl-2-butene (32 μL, 0.31 mmol) and sodium dihydrogen phosphate (37 mg, 0.31 mmol) in water (1.0 mL). To this solution was added [1-(2-{4-[2-(trifluoromethyl)phenoxy]piperidin-1-yl}pyrimidin-5-yl)-1H-1,2,3-triazol-4-yl]acetaldehyde (22 mg, 0.05 mmol) dissolved in acetone (1.0 mL). The reaction was stirred at room temperature for 1 h. The reaction was concentrated, poured into a 50-mL separatory funnel containing saturated aqueous KH2PO4 solution (30 mL) and the mixture was extracted with ethyl acetate (3×10 mL). The combined organic layers were washed with brine, dried over magnesium sulfate, filtered and the solvent was evaporated under reduced pressure to give the desired product.
