Reaction Steps

To a solution of paclitaxel (1.0 g, 1.17 mmol) in tetrahydrofuran (20 mL) was added hydrogen peroxide (30%, 20 mL) followed by sodium bicarbonate (1.92 g, 22.85 mmol). After stirring at room temperature for 18 hrs, the mixture was treated with dichloromethane/water (1:1(v/v), 100 mL). The organic phase was washed with water (2×30 mL), dried over anhydrous magnesium sulfate, and concentrated under vacuum. The residue was purified by silica gel flash chromatography eluting with 3% methanol/dichloromethane (v/v) to yield 10-deacetylpaclitaxel (890 mg, 93% yield) as a white powder.
