Reaction Conditions

Temperature-78 °C

Reaction Steps

Under an argon atmosphere and at −78° C., n-butyl lithium (a 1.57 M hexane solution, 701 μl, 1.10 mmol) was added to a dimethoxyethane solution (6 ml) of the 2-[(4-chlorophenyl)sulfonylmethyl]-1,4-difluorobenzene (303 mg, 1.00 mmol) obtained in Example 5. After the temperature of the reaction mixture was elevated to room temperature, it was cooled again to −78° C. After addition of 5-bromo-1-(1-pyrrolildinyl)-1-pentanone (281 mg, 1.20 mmol), the resulting mixture was stirred at room temperature for 20 hours. The reaction mixture was added with water, and then, diluted with dichloromethane. The solution thus obtained was washed successively with water and brine, dried over magnesium sulfate and then, concentrated. The residue thus obtained was subjected to flash chromatoraphy on a silica gel column, and the fraction obtained from the hexane:ethyl acetate=1:2 eluate was concentrated. The resulting solid was recrystallized from ethyl acetate-hexane, whereby the title compound (385 mg, 0.844 mmol, 84%) was obtained as a colorless solid.
