To a solution of racemic tert-butyl 4-(2-(2,6-dimethylphenyl)-6-(5-isopropyl-2-methylphenyl)-5,6,7,8-tetrahydropyrido[4,3-d]pyrimidin-4-yl)-(trans)-2,5-dimethylpiperazine-1-carboxylate (0.227 g, 0.389 mmol) and DCM (3 mL) at rt was added TFA (1 mL). After 25 min an additional 0.5 mL aliquot of TFA was added. After 1 h more the reaction was concentrated under reduced pressure to give 2-(2,6-dimethylphenyl)-4-((trans)-2,5-dimethylpiperazin-1-yl)-6-(5-isopropyl-2-methylphenyl)-5,6,7,8-tetrahydropyrido[4,3-d]pyrimidine as a TFA salt. MS (ESI+) m/z 484.4 (M+H)+
