7-(3,5-Dimethylisoxazol-4-yl)-6-methoxy-N4-(1-(1-methyl-1H-pyrazol-4-yl)ethyl)quinoline-3,4-diamine (70 mg, 0.178 mmol, for a preparation see intermediate 22) was dissolved in ethanol (5 ml) and ethylisothiocyanate (31.1 mg, 0.357 mmol) was added. The solution was heated at 60° C. for 3 h, then cooled and evaporated in vacuo. The residue was dissolved in THF and heated to 60° C. for 4 h, then evaporated in vacuo and the residue dissolved in DMSO and purified by MDAP (formic method. The product-containing fractions were evaporated in vacuo to give 7-(3,5-dimethylisoxazol-4-yl)-N-ethyl-8-methoxy-1-(1-(1-methyl-1H-pyrazol-4-yl)ethyl)-1H-imidazo[4,5-c]quinolin-2-amine (32 mg, 0.072 mmol, 40.3% yield) as a beige solid. LCMS (formate) Rt 0.71 min, MH+ not found.
