Reaction Steps
1M Aqueous sodium hydroxide solution (17.25 mL, 17.25 mmol) was added dropwise to a stirred solution of 6,7-dichloro-2,3-dimethoxy-5-[3-ethoxycarbonyl-5-(3-pyridyl)-4H-1,2,4-triazol-4-yl]quinoxaline (Preparation 94, 8.2 g, 17.25 mmol) in 1,4-dioxane (68 mL) and water (50 mL) at 10° C. The solution was warmed to room temperature and stirred for 20 hours, diluted with water (50 mL), acidified with glacial acetic acid and extracted with ethyl acetate (1×100 mL, 2×50 mL). The combined organic extracts were washed with brine, dried (MgSO4) and concentrated under reduced pressure. The residue was purified by flash chromatography on silica gel, by gradient elution using dichloromethane:methanol (1:0 changing to 9:1, by volume) to give the title compound (5.82 g, 84%) as a white solid, mp 206-207° C.
