Reaction Conditions

Temperature160 °C

Reaction Steps

3-Aminopicolinic acid (A-1) (1.0 g, 7.24 mmol, 1.0 eq), potassium cyanate (2.94 g, 36.2 mmol, 5.0 eq) and ammonium chloride (3.87 g, 72.4 mmol, 10.0 eq) are suspended in water (40 mL). The slurry is heated to 160° C. and mixed manually for 2 h as water vapour is expelled from the reaction vessel. The reaction temperature is then increased to 200° C. and stirred for 40 min. After cooling to 90° C., hot water is added and then the mixture is allowed to cool to RT. The mixture is acidified with concentrated HCl to adjust pH to 3-4. The precipitate is collected by filtration, rinsed with water (2×10 mL) and dried in vacuo to afford the product, pyrido[3,2-d]pyrimidine-2,4-diol (B-1).
