Para-Tolyl isocyanate (4.5 g 0.0339 mol) was added to a solution of 3-(4-aminophenyl)-5-({[(3-pyrrolidin-1-ylpropyl)amino]carbonyl}amino)isothiazole-4-carboxamide (12.0 g, 0.0308 mol), diisopropylethylamine (6.0 g, 0.046 mol) in mixture of tetrahydrofuran (300 mL) and dimethylformamide (50 ml) at ambient temperature over 20 min. The resulting mixture was stirred at ambient temperature for 2 hr. The reaction mixture was concentrated in vacuo to give off white solid, which was stirred in 20% tetrahydrofuran and ethyl acetate (200 mL) mixture, filtered the solid and washed with ethyl acetate the title compound as a white solid (15.8 g, 98%).
