Reaction Steps

Sodium hydride (60% dispersion in oil, 0.72 g, 18 mmol) was added to a solution of tert-butyl 4-(2-(6-bromopyridin-2-yl)hydrazono)piperidine-1-carboxylate (5.5 g, 15 mmol) in DMF (50 mL) at room temperature under N2, and the resulting solution was stirred for 20 min. Methyl iodide (1.0 mL, 17 mmol) was added to the solution, and the resulting solution was stirred for 2 h at ambient temperature. The mixture was quenched with H2O (100 mL) and extracted with EtOAc (4×50 mL). The combined organic extracts were concentrated to an orange oil under reduced pressure. The oil was diluted with EtOAc (100 mL) and washed with brine (7×50 mL). The organics were dried over Na2SO4, filtered, and concentrated to dryness under reduced pressure to provide the title compound (5.7 g, 99%) as an orange oil: 1H NMR (300 MHz, DMSO-d6) δ 7.45 (dd, J=8.1, 7.5 Hz, 1H), 6.91 (d, J=7.5 Hz, 1H), 6.67 (d, J=8.4 Hz, 1H), 3.56 (t, J=6.0 Hz, 2H), 3.44 (t, J=6.0 Hz, 2H), 3.09 (s, 3H), 2.53-2.46 (m, 4H, overlap with solvent), 1.42 (s, 9H).
