Reaction Steps

To a solution of methyl 4-[(2R)-2-({[(2R)-2-(6-amino-3-pyridinyl)-2-hydroxyethyl]amino}-methyl)-3,4-dihydro-2H-chromen-6-yl]benzoate (Example 381, 40 mg, 0.1 mmol) in THF (2 mL) and methanol (2 mL) was added 2 M aq. lithium hydroxide (0.5 mL, 1 mmol). The mixture was stirred at room temperature for 18 hours and then concentrated in vacuo. Prep. HPLC of the residue afforded 10.8 mg (28%) of the desired product. The product had: 1H NMR (CDCl3, δ): 7.96 (d, J=8.2 Hz, 2 H), 7.90 (d, J=2.2 Hz, 1 H), 7.49-7.56 (m, 3 H), 7.32-7.39 (m, 2 H), 6.82 (d, J=9.0 Hz, 1 H), 6.59 (d, J=8.6 Hz, 1 H), 4.70 (dd, J=8.3, 4.4 Hz, 1 H), 4.14-4.23 (m, 1 H), 2.75-3.02 (m, 6 H), 1.99-2.10 (m, 1 H), 1.70-1.85 (m, 1 H); mass spectroscopy gave m/z=420.2 [M+H]+ (calc'd exact mass for C24H25N3O4=419.2).
