Reaction Steps
Add sodium hydride (20.7 mg, 60% dispersion in mineral oil, 0.52 mmol) to a solution of 2-(6-chloro-pyridin-2-yl)-3-(4-fluoro-phenyl)-pyrazolo[1,5-a]pyridine (33.5 mg, 0.10 mmol) in DMF (2.5 mL) and methanol (2.5 mL) at room temperature. Stir the reaction mixture at room temperature for 4 hours and at 80° C. for 48 hours. Cool to room temperature and concentrate in vacuo. Add water (20 mL) and extract the aqueous solution with methylene chloride (3×20 mL). Dry the combined organic layers with sodium sulfate and concentrate. Purify by flash column chromatography (SiO2, 50% ethyl acetate/hexanes) to give the title compound 31 mg (97%) as an off-white solid.
