A toluene solution (10 ml) of 2-chloro-5-iodopyridine (67 mg, 0.27 mmol), bis-(4-methoxy-benzyl)-[5-(2-morpholin-4-yl-6,7-dihydro-5H-pyrrolo[2,3-d]pyrimidin-4-yl)-pyrimidin-2-yl]-amine (100 mg, 0.18 mmol), Pd2(dba)3.CHCl3 (19 mg, 0.018 mmol), BINAP (17 mg, 0.02 mmol) and cesium carbonate (181 mg, 0.55 mmol) was degassed under ultrasonic irradiation, followed by stirring at 100° C. for 14 hours under a nitrogen atmosphere. The reaction mixture was cooled to room temperature, and diluted with water, followed by extraction with ethyl acetate. The organic layer was washed with saturated aqueous sodium chloride solution, dried over magnesium sulfate, and subsequently magnesium sulfate was filtered off. The filtrate was concentrated under reduced pressure, and the residue was purified by silica gel column chromatography (hexane/ethyl acetate=3/1), to obtain {5-[7-(6-chloro-pyridin-3-yl)-2-morpholin-4-yl-6,7-dihydro-5H-pyrrolo[2,3-d]pyrimidin-4-yl]-pyrimidin-2-yl}-bis-(4-methoxy-benzyl)-amine as a yellow solid (43 mg, 36%).
