Reaction Steps
Tert-butyl (2-chloro-5-cyano-3-(3-oxopropyl)phenyl)carbamate (113 mg, 0.366 mmol) was taken up in THF (2 mL) and 1-methylpiperazine (0.081 mL, 0.732 mmol) and acetic acid (0.042 mL, 0.732 mmol) were added. The reaction was stirred for 10 min, and then sodium triacetoxyborohydride (233 mg, 1.098 mmol) was added. The reaction was stirred at room temperature for 1 h. The reaction mixture was quenched with saturated NaHCO3 and extracted with EtOAc. The organic layer was dried over Na2SO4, filtered, and concentrated. The material was purified by flash column chromatography (0-10% MeOHIDCM; 24 g column). Tert-butyl (2-chloro-5-cyano-3-(3-(4-methylpiperazin-1-yl)propyl)phenyl)carbamate (120 mg) was obtained as a colorless oil.
