To a solution of 17.6 mg (0.0346 mmol) of N-(4-{(1S,2S)-2-[(tert-butoxycarbonyl)amino]-1-[(dimethylamino)carbonyl]-3-[(3S)-3-fluoropyrrolidin-1-yl]-3-oxopropyl}phenyl)-N-methyl-4-methoxybenzamide from Step H in 1 mL of dichloromethane was added 1 mL of trifluoroacetic acid. After 1 h at ambient temperature, the solution was concentrated in vacuo. Purification by reverse-phase preparative HPLC (10% to 80% acetonitrile in water) afforded the title compound as a foaming white solid. LC/MS 471.2 (M+1).
