Reaction Steps

tert-Butyl N-[7-(tert-butyl)-2,3-dihydro-2,2,4-trimethyl-1-benzofuran-5-yl]carbamate (7.17 g, 21.5 mmol) was dissolved in N,N-dimethylformamide (72 ml), and sodium hydride (0.62 g, 25.8 mmol) which had been washed with hexane was added at 0° C. carefully. After stirring at room temperature for 30 minutes, 3-chloro-2-methyl-1-propene (2.53 ml, 25.8 mmol) and potassium iodide (0.36 g, 2.2 mmol) were added and the mixture was stirred at the same temperature for 1 hour. The reaction mixture was washed with saturated brine and dried over sodium sulfate, and the solvent was removed under reduced pressure. The residue was purified by a column chromatography on a silica gel (hexane:ethyl acetate=4:1). The resultant oil was dissolved in methanol (85 ml), combined with concentrated hydrochloric acid (8.5 ml), and stirred under argon atmosphere at 50° C. for 2 hours. After cooling to 0° C., the mixture was made weakly basic using a 3N aqueous solution of sodium hydroxide, and extracted with ethyl acetate. The organic layer was washed with saturated brine, dried over sodium sulfate, and the solvent was removed under reduced pressure, and then crystallized from a cold hexane to obtain 6.20 g of the title compound.
