Reaction Steps

To a solution of 4-(4-tertbutyldimethylsilyloxyanilino)-6-(2-bromo-4-methylanilino)pyrimidine (Reference Example 3, 396 mg) in tetrahydrofuiran (1 ml) under nitrogen atmosphere was added portion wise sodium hydride (60% dispersion in mineral oil, 58 mg). The solution was stirred for 30 minutes. Bromoacetonitrile (131 mg) was added and the reaction stirred for a further 1 hour. The solvent was evaporated and the residue purified by column chromatography eluting with DCM:MeOH:concentrated ammonia; 97.8:2:0.2 to give the title compound (158 mg, 37%) as a solid. M/z 524 (MH+).
