Deficiency of zinc may lead to growth retardation, hypogonadism in males, anorexia (possibly due to changes in taste and smell), depressed mental function, dermatitis, impaired wound-healing, suppressed immune function, diarrhea, and abnormal vitamin A metabolism with impaired night vision. /Zinc supplements/

Recommended intakes may be increased and/or supplementation may be necessary in the following conditions (based on documented zinc deficiency): alcoholism; burns; cirrhosis of the liver; diabetes mellitus; eating disorders - anorexia nervosa, bulimia; gastrectomy; genetic disorders - celiac, Crohn's, diarrhea, sprue, ulcerative colitis; intestinal parasitism; malabsorption syndromes associated with pancreatic insufficiency - pancreatic disease, cystic fibrosis; renal diseases - nephrotic syndrome, renal failure, uremia; short bowel syndrome; skin disorders - exfoliative dermatoses, psoriasis; prolonged stress; prolonged trauma. /Zinc supplements/

Recommended intakes for all vitamins and most minerals are increased during breast-feeding. /Zinc supplements/

Recommended intakes for all vitamins and most minerals are increased during pregnancy. Many physicians recommend that pregnant women receive multivitamin and mineral supplements, especially those pregnant women who do not consume an adequate diet and those in high-risk categories (i.e., women carrying more than one fetus, heavy cigarette smokers, and alcohol and drug abusers). /Zinc supplements/

Some unusual diets (e.g., reducing diets that drastically restrict food selection) may not supply minimum daily requirements of zinc. Supplementation may be necessary in patients receiving total parenteral nutrition (TPN) or undergoing rapid weight loss or in those with malnutrition, because of inadequate dietary intake. /Zinc supplements/

Protein BindingZinc is 70% protein bound in plasma, partially to serum albumin.
Pharmacodynamics · 1 Eintrag

Zinc is a cofactor in many enzymes and mediates a number of catalytic, structural, and regulatory roles in the body. It has a wide therapeutic index and long duration of action, as most zinc in the body is reabsorbed. Patients should be counselled regarding the risk of administration in patients with severe kidney dysfunction.

The CIR Expert Panel concluded that the following 27 zinc salts are safe in cosmetics in the present practices of use and concentration described in this safety assessment when formulated to be non-irritating...Zinc Chloride...

Safe for use in cosmetics, with qualifications

Anaemia results from the excessive absorption of zinc suppressing copper and iron absorption, most likely through competitive binding of intestinal mucosal cells. Unbalanced levels of copper and zinc binding to Cu,Zn-superoxide dismutase has been linked to amyotrophic lateral sclerosis (ALS). Stomach acid dissolves metallic zinc to give corrosive zinc chloride, which can cause damage to the stomach lining. Metal fume fever is thought to be an immune response to inhaled zinc. In preemployment and periodic physical examinations, special attention should be given to the skin and to the history of allergic dermatitis, as well as to exposed mucous membranes, the eyes, and the respiratory system. ... Smoking history should be known. Measurement of urinary zinc excretion may be useful.

The assessment of zinc exposure can be accomplished through measurement of zinc. The presence of excess zinc ... can indicate high exposure to zinc; however, no information was found in the literature regarding the accuracy of these levels in predicting possible health effects. Blood Reference Ranges: Normal - average level 1200 ug/dl; Exposed - not established; Toxic - not established. Serum or Plasma Reference Ranges: Normal - average levels 100 ug/dl; Exposed - not established; Toxic - not established. Urine Reference Ranges: Normal - average levels 0.5 mg/g creatinine or 150 to 1200 ug/24 hours; Exposed - urinary concentrations of 600 to 700 ug/l were found in workers exposed to zinc oxide at levels of 3 to 5 mg/cu m; Toxic - greater than 1200 ug/24 hours has been indicated as a toxic urinary level of zinc; however no information was located relating to the severity of symptoms seen with high urinary zinc levels. /Zinc/

Respiratory Symptom Questionnaires: Questionnaires have been published by the American Thoracic Society and the British Medical Research Council. These questionnaires have been found to be useful in identification of people with chronic bronchitis, however certain pulmonary function tests such as FEV1 have been found to be better predictors of chronic airflow obstruction. /Zinc/

Chest Radiography: This test is widely used for assessing pulmonary disease. Chest radiographs have been found to be useful for detection of early lung cancer in asymptomatic people, especially for detection of peripheral tumors such as adenocarcinomas. However, even though OSHA mandates this test for exposure to some toxicants such as asbestos, there are conflicting views on its efficacy in detection of pulmonary disease. /Zinc/

The ability of zinc chloride to induce morphological transformation in Balb/3T3 mouse cells (Cell transformation Assay) was evaluated in the presence of added metabolic activation by Aroclor-induced rat liver S-9 fraction. Based on preliminary clonal toxicity determinations (exposure time = 2hrs), zinc chloride, diluted with phosphate buffered saline, was tested at 200, 400, 600 or 800ug/ml in the presence of metabolic activation, with cell survival ranging from 83% to 1% relative to the solvent control. Treatments at 600 and 800ug/ml were too toxic and the cells failed to grow. Treatments at 200 and 400ug/ml produced significantly greater transformation frequencies (p < 0.05 and p < 0.01, respectively, Modified Poisson Distribution) relative to the solvent control.

The frequency of forward mutations was determined at the thymidine kinase (TK) locus of the mouse lymphoma L5178Y cell line exposed in vitro to zinc chloride in the presence of metabolic activation provided by Aroclor-induced rat liver S9 fraction. Mutagenic effects were observed in cultures after exposure to test article at concentrations ranging from 4.2 to 56 ug/plate, as indicated by 2 to 19.2 fold increases in mean mutant frequency compared to controls. The percent growth ranged from 95% to 10%, while in preliminary cytotoxicity assay, all cells were killed at 50 ug/ml.

The Agency has sufficient information on the human health effects of zinc salts and on it potential for causing effects in fish and wildlife and the environment when used to control moss and fungus growth in outdoor residential areas, on structures, in pressure-treated lumber and incorporated into fibers used in carpet. The Agency concludes that products containing zinc salts for these uses are eligible for reregistration. Only certain generic physical chemistry data studies on zinc salts still are needed as comfirmatory information. The Agency has determined that zinc salt containing products, labeled and used as specified in this Reregistration Eligibility Document will not pose unreasonable risks or adverse effects to humans or the environment.

As the federal pesticide law FIFRA directs, EPA is conducting a comprehensive review of older pesticides to consider their health and environmental effects and make decisions about their future use. Under this pesticide reregistration program, EPA examines health and safety data for pesticide active ingredients initially registered before November 1, 1984, and determines whether they are eligible for reregistration. In addition, all pesticides must meet the new safety standard of the Food Quality Protection Act of 1996. Pesticides for which EPA had not issued Registration Standards prior to the effective date of FIFRA, as amended in 1988, were divided into three lists based upon their potential for human exposure and other factors, with List B containing pesticides of greater concern and List D pesticides of less concern. Zinc chloride is found on List D. Case No: 4099; Pesticide type: fungicide, herbicide, antimicrobial; Case Status: RED Approved 09/92; OPP has made a decision that some/all uses of the pesticide are eligible for reregistration, as reflected in a Reregistration Eligibility Decision (RED) document.; Active ingredient (AI): Zinc chloride; Data Call-in (DCI) Date(s): 10/07/92; AI Status: OPP has completed a Reregistration Eligibility Decision (RED) document for the case/AI.

Zinc performs catalytic, structural, and regulatory roles in the body. Zinc is a component of approximately 3000 human proteins. Zinc is cytoprotective against reactive oxygen species mediated apoptosis through the action of metallothioneins. In a promyelocytic leukemia cell line, zinc enhances the up-regulation of A20 mRNA, which, via the TRAF pathway, decreases NF-kappaB activation, leading to decreased gene expression and generation of TNF-α, IL-1β, and IL-8. In patients with diarrhea, zinc restores mucosal barrier integrity, restores enterocyte brush-border enzyme activity, promotes the production of antibodies, and promotes the production of circulating lymphocytes against intestinal pathogens. Zinc also directly affects ion channels as a potassium channel blocker of cAMP-mediated chlorine secretion. Zinc deficiency decreases thymulin, inhibiting T-helper cell maturation and decreased Th-1 cytokines like IL-2. Decreased IL-2 decreases the activity of NK cells and CD8+ T cells. Zinc deficiency also leads to the generation of CD4+ T cells, decreased NF-κB activation, decreased phosphorylation of IκB, and decreased binding of NF-κB to DNA.

Hepatocytes were grown in monolayer culture for 22 hr and subsequently treated with zinc chloride (100 uM) for 24 hr which produced a 15-fold increase in metallothionein proteins (MT) concentration. Following pretreatment with zinc chloride, hepatocytes were treated with 20 or 40 uM silver nitrate for 24 hr, and cytotoxicity was then assessed by enzyme leakage and loss of intracellular potassium. The toxicity of silver was significantly less in the zinc-pretreated cells. Furthermore, although zinc pretreatment had no appreciable effect on the hepatocellular uptake of silver, it did alter the subcellular distribution, and silver accumulated more in the cytosol and less in the nuclear, mitochondrial, and microsomal fractions. Silver in the cytosol of control cells was bound primarily to high molecular weight proteins, whereas silver was mainly associated with MT in the zinc-pretreated cells. Zinc-induced MT in rat primary hepatocyte cultures protects against silver-induced cytotoxicity which results from the binding of silver to MT with a concomitant reduction of metal content in critical organelles and proteins.

Metabolism/Metabolites · 2 Einträge

Zinc chloride dissociates into ions _in vivo_ and does not undergo further metabolism.

Zinc can enter the body through the lungs, skin, and gastrointestinal tract. Intestinal absorption of zinc is controlled by zinc carrier protein CRIP. Zinc also binds to metallothioneins, which help prevent absorption of excess zinc. Zinc is widely distributed and found in all tissues and tissues fluids, concentrating in the liver, gastrointestinal tract, kidney, skin, lung, brain, heart, and pancreas. In the bloodstream zinc is found bound to carbonic anhydrase in erythrocytes, as well as bound to albumin, _2-macroglobulin, and amino acids in the the plasma. Albumin and amino acid bound zinc can diffuse across tissue membranes. Zinc is excreted in the urine and faeces. (L49)

Zinc is approximately 33% orally bioavailable in humans but bioavailability can vary between patients and depending on current zinc levels. Further data regarding the pharmacokinetics of zinc chloride are not readily available.

Zinc is predominantly eliminated in the feces. Gastrointestinal elimination of zinc is responsible for approximately half of all zinc elimination.

In one study of healthy patients, the clearance of zinc was found to be 0.63 ± 0.39 μg/min.

In humans subjected to low doses of (65)zinc chloride and whole-body measurements ... /it was/ estimated absorption to range from 58% to 77% in 5 controls, and from 16% to 42% in 3 patients suffering from acrodermatitis enterohepatica. The whole-body zinc retention was measured for 34 days ... One week after oral tracer dose of 2 ug zinc chloride to mice, the highest concentrations were found in bone tissue followed by liver and kidney.

The whole-body retention at 95 hr after intravascular injection of carrier-free (65)zinc chloride to mature male rats amounted to 78.3% of the dose.

In one study, 95 hr after injection of (65)zinc chloride to rats, 20.1% of the dose was recovered in feces and only 0.6% in urine ...

BeschreibungZinc chloride is a white deliquescent salt. It forms acidic solutions in water and in polar organic solvents such as ethanol, acetone, and ether. Anhydrous zinc chloride hydrolyzes with moisture to form hydrochloric acid. It also forms complex ions with water, ammonia, and some organic solvents.Zinc chloride reacts with sulphide to minimise release of H2S gas in waste treatment facilities. Zinc chloride 50% solution also serves as a high-quality mercerising agent for cotton. Zinc chloride is incompatible with strong oxidising agents, moisture, cyanides, sulphides, and potassium.

VerwendenZinc chloride is used as a wood preservative and in fireproofing timber. Other uses are as a deodorant in disinfecting fluids; in dental cements; in electroplating; in etching metals and glass; as flux for soldering; as a mordant in printing and dyeing textiles; in making dry batteries; in denaturing alcohols; in vulcanizing rubber; in manufacturing parchment; in making artificial silk; in making activated carbon and cold-water glues; and in refining petroleum. Also, zinc chloride is used as a dehydrating and condensing agent in organic syntheses. In medicine it is used as an astringent and antiseptic.

Definitionzinc chloride: A white crystalline compound, ZnCl2. The anhydrous salt, which is deliquescent, can be made by the action of hydrogen chloride gas on hot zinc; r.d. 2.9; m.p. 283°C; b.p. 732°C. It has a relatively low melting point and sublimes easily, indicating that it is a molecular compound rather than ionic. Various hydrates also exist. Zinc chloride is used as a catalyst, dehydrating agent, and Ûux for hard solder. It was once known as butter of zinc.
