Treatment · 1 Eintrag

Antidotes to cyanide poisoning include hydroxocobalamin and sodium nitrite, which release the cyanide from the cytochrome system, and rhodanase, which is an enzyme occurring naturally in mammals that combines serum cyanide with thiosulfate, producing comparatively harmless thiocyanate. Oxygen therapy can also be administered. (L97)

Target OrgansEyes, skin, respiratory system
Health Effects · 1 Eintrag

Exposure to high levels of cyanide for a short time harms the brain and heart and can even cause coma, seizures, apnea, cardiac arrest and death. Chronic inhalation of cyanide causes breathing difficulties, chest pain, vomiting, blood changes, headaches, and enlargement of the thyroid gland. Skin contact with cyanide salts can irritate and produce sores. (L96, L97)

Environmental Fate · 3 Einträge

TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 3.7(SRC), determined from a structure estimation method(2), indicates that methyl 2-cyanoacrylate is expected to have very high mobility in soil(SRC). However, movement of methyl 2-cyanoacrylate through soil will be hindered by its rapid polymerization as it comes into contact with moisture or other weakly basic substances(3-5). Since methyl 2-cyanoacrylate polymerizes rapidly on contact with moisture, volatilization from moist soil surfaces is not a relevant environmental process for this substance. Methyl 2-cyanoacrylate is not expected to volatilize from dry soil surfaces(SRC) based upon a vapor pressure of 0.2 mm Hg at 25 °C(3). Biodegradation is not an important environmental fate proces due to rapid polymerization(3-5).

AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 3.7(SRC), determined from a structure estimation method(2), indicates that methyl 2-cyanoacrylate is not expected to adsorb to suspended solids and sediment(SRC). However, methyl 2-cyanoacrylate polymerizes rapidly on contact with moisture or other weakly basic substances(3-5). Therefore, environmental processes such as volatilization from water surfaces and bioconcentration in aquatic organisms are not relevant for this substance(SRC). Biodegradation is not an important environmental fate proces due to rapid polymerization(3-5).

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), methyl 2-cyanoacrylate, which has a vapor pressure of 0.2 mm Hg at 25 °C(2), is expected to exist solely as a vapor in the ambient atmosphere. Vapor-phase methyl 2-cyanoacrylate is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 5.2 days(SRC), calculated from its rate constant of 3.1X10-12 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(3). Methyl 2-cyanoacrylate contains chromophores that absorb strongly at wavelengths >290 nm, and therefore may be susceptible to direct photolysis by sunlight(4).

Adverse EffectsSkin Sensitizer - An agent that can induce an allergic reaction in the skin.

Following sc implantation of (14)C-labeled methyl 2-cyanoacrylate into dogs radioactive urea was found in the urine. Hypothesis: hydrolytic chain scission & ester hydrolysis may be involved in in vivo degradation of cyanoacrylate tissue adhesives.

Organic nitriles are converted into cyanide ions through the action of cytochrome P450 enzymes in the liver. Cyanide is rapidly absorbed and distributed throughout the body. Cyanide is mainly metabolized into thiocyanate by either rhodanese or 3-mercaptopyruvate sulfur transferase. Cyanide metabolites are excreted in the urine. (L96)

Absorption, Distribution and Excretion · 4 Einträge

Following oral administration of 0.05 mL liquid beta-(14)C-methyl cyanoacrylate (MCA) (as monomer and polymer) to groups of two rats, radioactivity was detected in the urine, indicating that some absorption had occurred. For the rats receiving liquid MCA monomer into the oral cavity by syringe, approximately 2% of the radiolabel administered was recovered in urine over 48 hr. For rats receiving poly-MCA directly into the stomach, the total mean recovery from urine samples after 4 days was approximately 16%. A mean total of 18% of the radiolabel administered was recovered from feces on completion of the 4-day sampling period. However, the detection of radiolabel in the stool samples may well reflect the passage of poly-MCA particles directly through the gastrointestinal tract rather than any absorption and metabolism.

Following dermal application of 0.5 mL liquid liquid beta-(14)C-methyl cyanoacrylate (MCA) to groups of two rats, radioactivity was detected in the urine, indicating limited dermal absorption (up to 4% of the applied dose over a 6-day collection period). When the liquid was applied to skin with the epidermis removed, the amount of radiolabel recovered from urine increased approximately 3-fold.

Methyl 2-cyanoacrylate was rapidly absorbed from the skin incision site & eliminated in the urine in guinea pigs. Initial metabolites possessed the carbon skeleton of the monomer, whereas metabolites excreted from day 2 onward represented absorbed & degraded polymeric material.

The alkyl /cyanoacrylate/ esters can be absorbed through skin, with the methyl showing the highest rate of urine excretion, 12% of cmpd being excreted in five days when originally applied to rat skin.

The important step in most of the published syntheses is the classical Knoevenagel reaction using formaldehyde and a cyanoacetic ester. In this first step oligomeric cyanoacrylates, water, and other byproducts are formed. The reaction can be catalyzed by bases (e.g., amines), alkali metal tetraborates, metal carbonyls, or phase-transfer catalysts. The raw condensation product from the first reaction is depolymerized thermally (either continuously or discontinuously) and purified by distillation, chromatography on alumina or silica gel, crystallization in polyethylene columns, or treatment with zinc chloride. Alternatively, 2-cyanoacrylates can be manufactured by the ethoxycarbonylation of cyanoacetylene in the presence of nickel carbonyl. /2-Cyanoacrylates/

... /Alkyl 2-cyanoacrylates/ are prepared by pyrolyzing the poly(alkyl)-2-cyanoacrylates produced when formaldehyde is condensed with the corresponding alkyl cyanoacetates. /Cyanoacrylate adhesives/

Acrylate esters can be made by reacting a vinyl halide with carbon monoxide and an organic halide in the presence of a group VIII catalyst. Also developed is a process that yields acrylate esters after the generation of 2-halo-1-alkenes from hydrocarbon streams. Both processes were developed by Dow Chemical Co.

BeschreibungMethyl cyanoacrylate (MCA) is an organic compound that contains several functional groups, a methyl ester, a nitrile, and an alkene. It is a colorless liquid with low viscosity. Its chief use is as the main component of cyanoacrylate glues. It can be encountered under many trade names. Methyl cyanoacrylate is less commonly encountered than ethyl cyanoacrylate. It is soluble in acetone, methyl ethyl ketone, nitro methane, and dichloromethane. MCA polymerizes rapidly in presence of moisture.
