Versuchsvorschrift

A glass microwave reaction vessel was charged with tert-butyl 3-(6-chloropyrazin-2-yl)-5-(2,6-difluorophenyl)-1H-indole-1-carboxylate (50.0 mg, 0.113 mmol) and 4-boc-amino-1-piperidine (34.0 mg, 0.170 mmol) in NMP (0.6 mL), followed by Et3N (0.047 mL, 0.339 mmol). The reaction mixture was stirred and heated in an oil bath at 150° C. for 16 h, and then the mixture was diluted with DCM and water. The organic layer was separated, dried over Na2SO4, filtered and concentrated. The residue was dissolved in DCM (2 mL) and treated with TFA (0.5 mL). The reaction mixture was stirred at RT for 2 h, and solvent was removed. The residue was purified with RP-HPLC to give 1-(6-(5-(2,6-difluorophenyl)-1H-indol-3-yl)pyrazin-2-yl)piperidin-4-amine as a brown solid. MS (ESI, pos. ion) m/z: 406 (M+1).
