Overview

2-dimethylaminoethyl methacrylate appears as a clear colorless liquid. Less dense than water and insoluble in water. Vapors heavier than air and corrosive to eyes and mucous membranes. May polymerize exothermically if heated or contaminated. If polymerization takes place inside a closed container, the container may rupture violently. Produces toxic oxides of nitrogen during combustion. Toxic by skin absorption, ingestion and inhalation. Used to make plastics and in textiles.

Environmental Fate · 3 items

TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 20(SRC), determined from a structure estimation method(2), indicates that N,N-dimethylaminoethyl methacrylate is expected to have very high mobility in soil(SRC). The pKa of N,N-dimethylaminoethyl methacrylate is 8.44(3), indicating that this compound will exist mostly in cation form in the environment and cations generally adsorb more strongly to soils containing organic carbon and clay than their neutral counterparts(4). Volatilization of N,N-dimethylaminoethyl methacrylate from moist soil surfaces is not expected to be an important fate process(SRC) given an estimated Henry's Law constant of 1.4X10-7 atm-cu m/mole(SRC), using a fragment constant estimation method(2). N,N-Dimethylaminoethyl methacrylate is expected to volatilize from dry soil surfaces(SRC) based upon an estimated vapor pressure of 0.72 mm Hg at 25 °C(SRC), determined from a fragment constant method(2). Using the OECD 301E test, 95.3% degradation was reached in 2 weeks(5) suggesting that biodegradation is an important environmental fate process in soil(SRC). Based on results of the Japanese MITI test which noted abiotic hydrolysis during biodegradation studies(5,6), N,N-dimethylaminoethyl methacrylate may degrade via abiotic hydrolysis in moist alkaline and neutral soils(SRC).

AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 20(SRC), determined from a structure estimation method(2), indicates that N,N-dimethylaminoethyl methacrylate is not expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is not expected(3) based upon an estimated Henry's Law constant of 1.4X10-7 atm-cu m/mole(SRC), developed using a fragment constant estimation method(2). According to a classification scheme(4), an estimated BCF of 2.6(SRC), from a log Kow of 1.13(5) and a regression-derived equation(2), suggests the potential for bioconcentration in aquatic organisms is low(SRC). Using the OECD 301E test, 95.3% degradation was reached in 2 weeks(5) suggesting that biodegradation is an important environmental fate process in water(SRC). The hydrolysis half-life of N,N-dimethylaminoethyl methacrylate at 25 °C was determined to be 4.54 days and 3.31 hours at pH 7 and pH 9 respectively(5); the compound is stable to hydrolysis at pH 4(5). N,N-Dimethylaminoethyl methacrylate is an olefin and olefins in surface waters exposed to sunlight have a half-life on of 25 days(6) .

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), N,N-dimethylaminoethyl methacrylate, which has an estimated vapor pressure of 0.72 mm Hg at 25 °C(SRC), determined from a fragment constant method(2), is expected to exist solely as a vapor in the ambient atmosphere. Vapor-phase N,N-dimethylaminoethyl methacrylate is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 3.9 hours(SRC), calculated from its rate constant of 9.9X10-11 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(2). Vapor-phase N,N-dimethylaminoethyl methacrylate is also degraded in the atmosphere by reaction with ozone(SRC); the half-life for this reaction in air is estimated to be 24 hours(SRC), calculated from its rate constant of 1.1X10-17 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(2). Based on analogy to a measured nitrate radical rate constant of 4.90X10-15 cu cm/molecule-sec at 25 °C for ethyl methacrylate(3), the atmospheric half-life of N,N-dimethylaminoethyl methacrylate is estimated to be about 6.5 days(SRC).

Gene transfection is the transfer of genetic material like DNA into cells. Cationic polymers which form nanocomplexes with DNA, so-called non-viral gene vectors, are a highly promising platform for efficient gene transfection. Despite intensive research efforts and some of the on-going clinical trials on gene transfection, none of the existing cationic polymer systems are generally acceptable for human gene therapy. Since the process of gene transfection is complex and puts different challenges and demands on the delivery system, there is a strong requirement for the design and development of a multifunctional system in a simple way. This review will discuss recent efforts in design, synthesis, and performance of poly(2-dimethylaminoethyl methacrylate) (PDMAEMA) nanocomplexes with DNA.

Applications · 1 item

UsesPoly(2-(dimethylamino)ethyl methacrylate) is a pH-responsive polymer. pH-responsive polymers are a group of stimuli-responsive polymers th at can respond to solution pH by undergoing structural and property changes such as surface activity, chain conformation, solubility, and configuration. The term “pH-responsive polymers” is commonly used to describe polymers having ionisable acidic or basic residues whose ionization depends on solution pH. The physical properties of the polymer, such as its chain conformation, configuration, and solubility, can be tailored by manipulating the pH or ionic strength. These unique properties of pH responsive polymer systems consequently make them very useful in various applications such as drug delivery, gene delivery, sensors, surfaces, membranes, and chromatography.pH-sensitive polymer systems combined with nanotechnology could be utilized as an alternative strategy to traditional targeting systems to overcome major problems in current chemotherapy represented by non-specific tissue distribution of the drugs, tumor heterogeneity, and multidrug resistance (MDR) against anticancer drugs.
