Uses & manufacturing
Applications
Uses2-Amino-2',5-dichlorobenzophenone is used to produce lorazepam a drug of the benzodiazepine group, used especially to treat anxiety.
UsesLorazepam Impurity A.
General Description2-Amino-2′,5-dichlorobenzophenone can be synthesized from a precursor diazepine, Iorazepam.
Preparation methods
PreparationAt room temperature, 2-amino-5-chlorobenzonitrile (0.4 mmol), 2-chlorophenylboronic acid (0.8 mmol), Pd(O2CCF3)2 (5 mol%), MsOH (10 eq), ligand 5,5'-dimethyl-2,2'-bipyridine (7.5 mol%), 2-methyltetrahydrofuran (2 mL), and H2O (1 mL) were added to a Schlenk tube. The reaction mixture was stirred vigorously at 80 °C for 36 h. The mixture was poured into ethyl acetate, washed with saturated NaHCO3 (2 × 10 mL), and then washed with brine (1 × 10 mL). The aqueous layer was extracted with ethyl acetate, the organic layers were combined and dried over anhydrous Na2SO4, and concentrated under vacuum. The residue was purified by flash column chromatography (using hexane/ethyl acetate as eluent) to give the desired yellow solid product 2-amino-2',5-dichlorobenzophenone (55.3 mg, 52% yield).
