Pharmacology

Metabolism/Metabolites · 1 item

Arsenic is absorbed mainly by inhalation or ingestion, as to a lesser extent, dermal exposure. It is then distributed throughout the body, where it is reduced into arsenite if necessary, then methylated into monomethylarsenic (MMA) and dimethylarsenic acid (DMA) by arsenite methyltransferase. Arsenic and its metabolites are primarily excreted in the urine. Arsenic is known to induce the metal-binding protein metallothionein, which decreases the toxic effects of arsenic and other metals by binding them and making them biologically inactive, as well as acting as an antioxidant. (L20)

PIGS FED WITH ARSANILIC ACID HAD HIGH LEVELS (OVER 20 PPM) OF ARSENIC IN LIVER & KIDNEY CORTEX AFTER FEW DAYS OF ILLNESS, LEVELS THEN DECR ALTHOUGH ADMIN OF DRUG CONTINUED...

Experiments with pigs given a diet containing 0.01% arsanilic acid for 31 days showed a rapid decr in the tissue levels of arsenic during the first days after the feeding of arsanilic acid was discontinued. The livers contained arsenic concns of 1.5-2 mg/kg on the 31st day of feeding & about 0.2 mg/kg on the 7th day after the removal of the arsanilic acid from the diet.

Rats fed protein from swine liver containing 24.4 mg arsenic/kg protein as arsanilic acid, at a level of 300 g/kg diet for 14 days, eliminated almost all of the arsenic within 7 days of the end of the feeding period. The amount of arsenic excreted in the urine was one third of the amount of arsenic eliminated in the feces during this period.

...FED ARSANILIC ACID TO SWINE...5-10% OF AS EXCRETED IN FECES WAS ARSANILIC ACID. A RAPID DECR IN TISSUE AS & EXCRETION FOLLOWING REMOVAL OF ADDITIVE WAS FOUND. @ HIGH RATES, AS ALSO APPEARED IN URINE, BUT NOT AS ARSANILIC ACID.

For the maximum protection of human health from the potential carcinogenic effects due to exposure of arsenic through ingestion of contaminated water & contaminated aquatic organisms, the ambient water concn should be zero based on the nonthreshold assumption for this chemical. However, zero level may not be attainable at the present time. Therefore, the levels which may result in incremental increase of cancer risk over the lifetime are estimated at 1X10-5, 1X10-6, & 1X10-7. The corresponding criteria are 22 ng/l, 2.2 ng/l, & .22 ng/l, respectively .... For consumption of aquatic organisms only, excluding consumption of water, the levels are 175 ng/ml, 17.5 ng/l, & 1.75 ng/l, respectively. /Arsenic/

D004; A solid waste containing arsenic may or may not become characterized as a hazardous waste when subjected to the Toxicity Characteristic Leaching Procedure listed in 40 CFR 261.24, and if so characterized, must be managed as a hazardous waste.

Toxicology

Treatment · 1 item

Arsenic poisoning can be treated by chelation therapy, using chelating agents such as dimercaprol, EDTA or DMSA. Charcoal tablets may also be used for less severe cases. In addition, maintaining a diet high in sulfur helps eliminate arsenic from the body. (L20)

Interactions · 1 item

... The effects of cadmium (50 mg/kg diet) & lead (200 mg/kg) on the toxicity of arsenic administered to rats as sodium arsenate or arsanilic acid in the food (50 mg As/kg diet) /was examined/. The efficiency of food utilization was more reduced by the combination of arsenic & cadmium than by each metal alone. The combination of arsenic & cadmium also caused a greater decr in serum alkaline phosphatase levels than either metal alone. Additive effects of arsenic & lead in coproporphyrin excretion were also noted. Increased uroporphyrin excretion in rats was reported ..., when lead cadmium & arsenic were combined compared with excretion levels following exposure to arsenic alone.

Health EffectsArsenic poisoning can lead to death from multi-system organ failure, probably from necrotic cell death, not apoptosis. Arsenic is also a known carcinogen, especially in skin, liver, bladder and lung cancers. (T1, L20)
Adverse EffectsOccupational hepatotoxin - Secondary hepatotoxins: the potential for toxic effect in the occupational setting is based on cases of poisoning by human ingestion or animal experimentation.
Exposure RoutesOral (L2) ; inhalation (L2); dermal (L2)
Toxicity Summary · 1 item

Arsenic and its metabolites disrupt ATP production through several mechanisms. At the level of the citric acid cycle, arsenic inhibits pyruvate dehydrogenase and by competing with phosphate it uncouples oxidative phosphorylation, thus inhibiting energy-linked reduction of NAD+, mitochondrial respiration, and ATP synthesis. Hydrogen peroxide production is also increased, which might form reactive oxygen species and oxidative stress. Arsenic's carcinogenicity is influenced by the arsenical binding of tubulin, which results in aneuploidy, polyploidy and mitotic arrests. The binding of other arsenic protein targets may also cause altered DNA repair enzyme activity, altered DNA methylation patterns and cell proliferation. (T1, A17)

Signs and Symptoms · 1 item

Exposure to lower levels of arsenic can cause nausea and vomiting, decreased production of red and white blood cells, abnormal heart rhythm, damage to blood vessels, and a sensation of “pins and needles” in hands and feet. Breathing high levels of inorganic arsenic can give you a sore throat or irritated lungs. Arsenic also affects the brain, causing neurological disturbances such as headaches, confusion, and drowsiness. (A1)

UsesArsanilic acid reacts essentially analogously to phenylarsonic acid. According to Chandelle, in the presence of relatively large amounts of accompanying ions the determination of zirconium gives more accurate results with this reagent than with phenylarsonic acid. This statement has not yet been confirmed. Nevertheless, the reagent is much more widely used than any other arsonic acid derivative.

Uses4-Aminophenylarsonic acid is an organoarsenic compound. 4-Aminophenylarsonic acid is a highly toxic contaminant and can be found in plants growing in contaminated soil. 4-Aminophenylarsonic acid was used as an additive in animal feed.
