Environmental Fate
TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 74(SRC), determined from a log Kow of 1.83(2) and a regression-derived equation(3), indicates that methyl phenylacetate is expected to have high mobility in soil(SRC). Volatilization of methyl phenylacetate from moist soil surfaces may be an important fate process(SRC) given an estimated Henry's Law constant of 1.4X10-5 atm-cu m/mole(SRC), using a fragment constant estimation method(4). Methyl phenylacetate has a vapor pressure of 0.13 mm Hg(5) and exists as a liquid at environmental standard temperature and pressure; therefore, methyl phenylacetate may volatilize from dry soil(SRC). Utilizing the Closed Bottle test, 75% degradation was reached in 28 days(5) indicating that biodegradation is an important environmental fate process in soil(SRC).
AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 74(SRC), determined from a log Kow of 1.83(2) and a regression-derived equation(3), indicates that methyl phenylacetate is not expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is expected(4) based upon an estimated Henry's Law constant of 1.4X10-5 atm-cu m/mole(SRC), developed using a fragment constant estimation method(5). Using this Henry's Law constant and an estimation method(4), volatilization half-lives for a model river and model lake are 2 and 28 days, respectively(SRC). According to a classification scheme(6), an estimated BCF of 7.5(SRC), from its log Kow(2) and a regression-derived equation(3), suggests the potential for bioconcentration in aquatic organisms is low(SRC). Utilizing the Closed Bottle test, 75% degradation was reached in 28 days(7) indicating that biodegradation is an important environmental fate process in water(SRC).
ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), methyl phenylacetate, which has a vapor pressure of 0.13 mm Hg at 20 °C(2), is expected to exist solely as a vapor in the ambient atmosphere. Vapor-phase methyl phenylacetate is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 8.6 days(SRC), calculated from its rate constant of 3.7X10-12 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(3). Methyl phenylacetate does not absorb UV light at wavelengths >290 nm(4) and, therefore, is not expected to be susceptible to direct photolysis by sunlight(SRC).
Toxicity Summary
IDENTIFICATION AND USE: Methyl phenyl acetate is used as food and beverage ingredient. It is also the starting material in manufacture of synthetic perfumes. HUMAN STUDIES: There are no data available. ANIMAL STUDIES: Restrained bees were tested with methyl phenylacetate previously identified from Mycobacterium tuberculosis cultures, to determine honeybee capacity for signature volatile detection. Methyl phenylacetate was detectable over eight orders of magnitude. Potential exists for trained honeybees in non-invasive diagnostic tests for TB.

Artificial Pollution Sources
Methyl phenylacetate's production and use as a synthetic food flavoring agent(1) may result in its release to the environment through various waste streams(SRC). Its use in the manufacture of synthetic perfumes(2) may result in its direct release to the environment(SRC).
Environmental Bioconcentration
An estimated BCF of 7.5 was calculated in fish for methyl phenylacetate(SRC), using a log Kow of 1.83(1) and a regression-derived equation(2). According to a classification scheme(2), this BCF suggests the potential for bioconcentration in aquatic organisms is low(SRC).
Volatilization from Water/Soil
The Henry's Law constant for methyl phenylacetate is estimated as 1.4X10-5 atm-cu m/mole(SRC) using a fragment constant estimation method(1). This Henry's Law constant indicates that methyl phenylacetate is expected to volatilize from water surfaces(2). Based on this Henry's Law constant, the volatilization half-life from a model river (1 m deep, flowing 1 m/sec, wind velocity of 3 m/sec)(3) is estimated as 2 days(SRC). The volatilization half-life from a model lake (1 m deep, flowing 0.05 m/sec, wind velocity of 0.5 m/sec)(3) is estimated as 28 days(SRC). Methyl phenylacetate's Henry's Law constant indicates that volatilization from moist soil surfaces may occur(SRC). Methyl phenylacetate has a vapor pressure of 0.13 mm Hg(4) and exists as a liquid at environmental standard temperature and pressure; therefore, methyl phenylacetate may volatilize from dry soil(SRC).
Non-Human Toxicity Excerpts
/OTHER TOXICITY INFORMATION/ The proboscis extension reflex in honeybees was evaluated for detection of tuberculosis. Restrained bees were tested with methyl phenylacetate, methyl p-anisate, and methyl nicotinate, previously identified from Mycobacterium tuberculosis cultures, to determine honeybee capacity for signature volatile detection. Methyl p-anisate and methyl phenylacetate were detectable over eight orders of magnitude, and honeybees showed proboscis extension response down to 0.1 pg loading of methyl p-anisate on filter paper. Potential exists for trained honeybees in non-invasive diagnostic tests for TB.

Metabolism/Metabolites
Under the auspices of the International Organization of the Flavor Industry (IOFI), preliminary studies were designed to establish the relative rates of enzymic hydrolysis of 26 esters of organic alcohols and acids of various structural configurations (iso-amyl furylpropanoate, ethyl cyclohexylpropanoate, allyl hexanoate, benzyl 2-methylbutanoate, m-cresyl phenylacetate, citronellyl acetate, benzylphenylacetate, ethylfurylpropanoate, allylphenylacetate, isoamylphenylacetate, ethyldecanoate, benzylcinnamate, methylphenylacetate, o-cresyl acetate, citronellylphenylacetate, benzylacetate, isopropylphenylacetate, isopropylbutanoate, isoamylacetate, 1,3-dimethylbutyl acetate, ethylacetate, benzyltiglate, allyltiglate, methylanthranilate, methyl-N-methylanthranilate and dimethylbenzylcarbinylacetate). In the majority of cases, the selected esters were hydrolyzed rapidly by pancreatin. Additional investigations were undertaken on 8 esters (isoamylacetate, ethylacetate, allyltiglate, 1,3-dimethylbutylacetate, benzyltiglate, methyl-N-methylanthranilate, methylanthranilate and dimethylbenzylcarbinyl acetate) not rapidly hydrolyzed using whole homogenates of pig jejunum and liver. There was wide variation in the degree of in vitro hydrolysis of flavoring esters depending on the source of enzymes used. Those esters hydrolyzed rapidly in vitro will possibly also be degraded readily in the intact animal.

調製方法
製造方法Methyl phenylacetate is synthesized from phenylacetonitrile by hydrolysis and esterification. Put methanol into a dry glass-lined reaction pot, stir and cool to below 30°C, add sulfuric acid dropwise, heat up to 90°C after adding, and start adding phenylacetonitrile dropwise, control the temperature at about 95°C, and finish adding in 1.5h. After reacting at 95-100 °C for 6 h, it was cooled to below 40 °C and diluted with water equivalent to about 0.6 times the reaction solution. The acid water was separated by standing, and a saturated sodium carbonate solution was added for neutralization and washing, and the aqueous layer was discarded. It is dehydrated with anhydrous calcium chloride and fractionated under reduced pressure to obtain methyl phenylacetate with a yield of 80%.
