Leaf Alcohol is a colorless liquid with the characteristic odor of freshly cut grass. In small quantities, leaf alcohol occurs in the green parts of nearly all plants. The volatile flavor constituents of green tea contain up to 30%. A stereospecific synthesis of (Z)-3-hexen-1-ol starts with the ethylation of sodium acetylide to 1-butyne, which is reacted with ethylene oxide to give 3-hexyn-1-ol. Selective hydrogenation of the triple bond in the presence of palladium catalysts yields (Z)-3-hexen-1-ol. Biotechnological processes have been developed for its synthesis as a natural flavor compound, for example. Leaf alcohol is used to obtain natural green top notes in perfumes and flavors. In addition, it is the starting material for the synthesis of (2E,6Z)-2,6-nonadien-l-ol and (2E,6Z)-2,6-nonadien-l-al.

Environmental Fate · 3건

TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 12(SRC), determined from a structure estimation method(2), indicates that cis-3-hexen-1-ol is expected to have very high mobility in soil(SRC). Volatilization of cis-3-hexen-1-ol from moist soil surfaces is expected to be an important fate process(SRC) given an estimated Henry's Law constant of 1.6X10-5 atm-cu m/mole(SRC), using a fragment constant estimation method(2). cis-3-Hexen-1-ol is expected to volatilize from dry soil surfaces(SRC) based upon an estimated vapor pressure of 0.94 mm Hg at 25 °C(SRC), determined from a fragment constant method(2). An 87% of theoretical BOD using activated sludge in the Japanese MITI test(3) suggests that biodegradation is an important environmental fate process in soil(SRC). cis-3-Hexen-1-ol absorbs at wavelengths >290 nm(4) and, therefore, may be susceptible to direct photolysis on soil surfaces exposed to sunlight(SRC).

AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 12(SRC), determined from a structure estimation method(2), indicates that cis-3-hexen-1-ol is not expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is expected(3) based upon an estimated Henry's Law constant of 1.6X10-5 atm-cu m/mole(SRC), developed using a fragment constant estimation method(2). Using this Henry's Law constant and an estimation method(3), volatilization half-lives for a model river and model lake are 2.5 and 21 days, respectively(SRC). According to a classification scheme(4), an estimated BCF of 5(SRC), from an estimated log Kow of 1.61(2) and a regression-derived equation(2), suggests the potential for bioconcentration in aquatic organisms is low(SRC). An 87% of theoretical BOD using activated sludge in the Japanese MITI test(5) suggests that biodegradation is an important environmental fate process in water(SRC). cis-3-Hexen-1-ol is not expected to undergo hydrolysis in the environment due to the lack of functional groups that hydrolyze under environmental conditions(3). cis-3-Hexen-1-ol is an olefin and olefins in surface waters exposed to sunlight react with photo-oxidants (such as hydroxyl radicals, peroxy radicals and singlet oxygen) with a half-life on the order of 25 days reported(6).

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), cis-3-hexen-1-ol, which has an estimated vapor pressure of 0.94 mm Hg at 25 °C(SRC), determined from a fragment constant method(2), is expected to exist solely as a vapor in the ambient atmosphere. Vapor-phase cis-3-hexen-1-ol is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 3.6 hours(SRC), calculated from its rate constant of 1.06X10-10 cu cm/molecule-sec at 24 °C(3). cis-3-Hexen-1-ol is also degraded in the atmosphere by reaction with ozone, nitrate radicals and direct photolysis(4). The half-life for the reaction with ozone is estimated to be 7.5 hours(SRC), calculated from its rate constant of 3.69X10-17 cu cm/molecule-sec at 25 °C(5). The half-life for the reaction with nitrate radicals is estimated to be 2.1 hours(SRC), calculated from its rate constant of 2.67X10-13 cu cm/molecule-sec at 25 °C(5). The nitrate radical (NO3) is the dominant atmospheric oxidant during the night-time in most atmospheric environments(6), therefore, night-time degradation appears to be a major fate process for cis-3-hexen-1-ol(SRC). cis-3-Hexen-1-ol absorbs at wavelengths >290 nm(4), and a calculated upper limit direct photolysis rate constant at ground level of 1.61X10-5/sec(4) corresponds to a half-life of about 12 hours(SRC).

Toxicity Summary · 1건

IDENTIFICATION AND USE: 3-Hexenol is a colorless liquid. It has a strong, fresh-cut grass odor and a fresh green, fruity taste. It is released to the air from a variety of plants including grass, clover, alfalfa, grapes, onions, peaches and oak trees. It is present in the green parts of nearly all plants. It is reported to be found in several essential oils and in many fruit juices and is a component of tobacco smoke. 3-Hexenol is used as a fragrance in perfumes, household cleaners, and food flavoring. HUMAN EXPOSURE AND TOXICITY: Tested at 4% in petrolatum, 3-hexenol produced no irritation after a 48 hour closed patch test in humans. Odorization of inert gas can serve to warn workers in an enclosed space. A psychophysical investigation examined 3-hexenol as a candidate for the possible odorization of argon. The detection threshold for 3-hexenol in argon was 19 ppb. 3-Hexenol did not induce any statistically significant increases in the frequency of human lymphocytes with chromosome aberrations in either the absence or presence of a liver enzyme metabolizing system. ANIMAL STUDIES: The LD50 of 3-hexenol in rats and mice was in the range of 7-10 g/kg by the oral route and 0.4-0.6 g/kg by the intraperitoneal route. No effects were seen at 310 and 1250 ppm, but in males of the 5000-ppm group the intake of drinking water was reduced compared with controls, the relative kidney weight was increased and the urine collected in the first 2 hr after a water load was more concentrated. In females, the only finding was a transitory anemia at this level. In another study, 3-hexanol applied at full strength to rabbit skin for 24 hours produced no irritation. In rats, an analysis of Fos protein was performed as a way of investigating the effects of inhalation of green odor (a mixture of of trans-2-hexenal and 3-hexenol) on the neuronal activations in stress-related forebrain regions induced by acute and repeated stress. Green odor had inhibitory effects on the stress-induced corticosterone response, body-weight loss, and adrenal hypertrophy. In another study, the reproductive toxicity via the oral route was assessed in rats at dose levels of 100, 300 and 1000 mg/kg/day. Based on the results of the study, a NOAEL for general toxicity in males and females was considered to be 1000 mg/kg/day, the highest dose level tested. The potential mutagenicity of 3-hexenol on the thymidine kinase, TK +/-, locus of the L5178Y mouse lymphoma cell line was assessed. 3-Hexenol did not induce any toxicologically significant increases in the mutant frequency at the TK +/- locus in L5178Y cells and is therefore considered to be non-mutagenic under the conditions of the test.

In response to insect feeding, corn plants (Zea mays cv. Delprim) release elevated levels of volatile organic compounds (VOCs), including the C(6)-volatile (Z)-3-hexenol. The level of mRNA accumulation for a series of defense genes was monitored in response to application of (Z)-3-hexenol (50 nmol) to undamaged plants. The induction of transcripts for hpl (hydroperoxide lyase), fps (farnesyl pyrophosphate synthase), pal (phenylalanine ammonia-lyase), lox (lipoxygenase), igl (indole-3-glycerol phosphate lyase) and mpi (maize proteinase inhibitor) were compared with metabolites generated from the respective pathways. While headspace VOC analysis showed an increase in (Z)-3-hexenyl acetate and methyl salicylate with lox and pal induction, respectively, MPI accumulation was not observed with an increase in mpi transcripts. Moreover, (Z)-3-hexenol treatment did not elevate sesquiterpene emissions or activate fps transcription. Chemical labeling and bioassay experiments established that exogenous (Z)-3-hexenol can be taken up and converted to a less active acetylated form. These data indicate that (Z)-3-hexenol can serve as a signaling molecule that triggers defense responses in maize and can rapidly be turned over in planta.
