실험 절차

(S)-4-((1-(5-Methyl-4-oxo-3-phenyl-3,4-dihydropyrrolo[2,1-f][1,2,4]triazin-2-yl)ethyl)amino)-7H-pyrrolo[2,3-d]pyrimidine-5-carboxylic acid (50 mg, 0.12 mmol), (1H-pyrazol-4-yl)methanamine (11 mg, 0.11 mmol) and diisopropylethylamine (163 μl, 0.94 mmol) were dissolved in DMF (2 ml) and stirred for 30 min. The reaction flask was then sealed and T3P® (50% in DMF, 270 μl, 0.46 mmol) was added dropwise and the reaction mixture stirred overnight at room temperature. The mixture was poured into ice-water and extracted with ethyl acetate. The resulting organic solution was washed with water and brine, dried over magnesium sulphate, filtered and the solvents were removed under reduced pressure. The product was purified by flash chromatography using SP1® Purification System (0% to 10% DCM/MeOH) to give 1.5 mg (2% yield) of the title compound as a white solid.
