실험 절차

A solution of (1R,10R,11S,16S)-5-methoxy-1,11,15,15-tetramethyl-8-thiatetracyclo[8.8.0.02,7.011,16]octadeca-2,4,6-triene-3-carboxylic acid (74) (0.13 g, 0.33 mmol) in anhydrous THF (10 mL) and anhydrous DMF (0.7 mL), was prepared under N2. To the solution, HATU (0.15 g, 0.40 mmol) and Hunig's base (0.08 mL, 0.4 mmol) were added. The mixture was stirred at room temperature for 1 h. MeNH2 (2 M in THF, 2.0 mL, 4.0 mmol) was added and the flask was sealed and stirred at room temperature for 24 h. The reaction mixture was concentrated, and the residue was dissolved in CH2Cl2 and washed with saturated aqueous sodium bicarbonate. The aqueous phase was extracted with CH2Cl2. The combined organic extracts were washed with brine and concentrated to dryness. The crude product was purified by flash chromatography on silica gel (CH2Cl2/EtOAc, 5:1) to yield (1R,10R,11S,16S)-5-methoxy-N,1,11,15,15-pentamethyl-8-thiatetracyclo[8.8.0.02,7.011,16]octadeca-2,4,6-triene-3-carboxamide (75) (0.08 g, 59%) as a white solid.
