A solution of tert-butyl (6-Methylamino-pyrimidin-4-yl)-carbamate (240 mg, 1.07 mmol, 1.0 eq.), commercially available 2-methyl-5-nitrophenylisocyanate (210 mg, 1.18 mmol, 1.1 eq.), and DMAP (26 mg, 0.21 mmol, 0.2 eq.) in toluene (10 ml) is stirred at 80° C. for 24 h. After cooling to room temperature methanol (10 ml) is added and the suspension formed is stirred for 10 min at 50° C. The precipitate is filtered off and washed with methanol (2×10 ml). After drying in vacuo an extremely insoluble colorless powder is obtained: HPLC: tR=8.09 min (purity: 85%, gradient A), ESI-MS: 403.5 [MH]+.
