A 100 mL round bottom flask was charged with 5-bromo-2-methyl-8-(piperazin-1-yl)-[1,2,4]-triazolo[1,5-a]pyrazine (0.5 g, 1.7 mmol), CH2Cl2 (20 mL), MeOH (10 mL), 40% aqueous formaldehyde (2 mL) and NaBH3(CN) (0.5 g, 8.0 mmol). The resulting solution was stirred at room temperature for 0.5 h. Work-up: the reaction mixture was poured into saturated aqueous NaHCO3 and extracted with CH2Cl2 (50 mL×3). The combined organic layers were dried over anhydrous Na2SO4 and concentrated in vacuo. The residue was purified by flash column chromatography on silica gel with 1-10% MeOH in CH2Cl2, to afford 0.4 g (76%) of the product as a yellow solid. MS m/z: 311 (M+H+).
