Procedure

3-Aminopicolinic acid (B-1) (1.0 g, 7.24 mmol, 1.0 eq), potassium cyanate (2.94 g, 36.2 mmol, 5.0 eq) and ammonium chloride (3.87 g, 72.4 mmol, 10.0 eq) were suspended in water (40 mL). The slurry was heated to 160° C. and mixed manually for 2 h as water vapor was expelled from the reaction vessel. The reaction temperature was then increased to 200° C. and stirred for 40 min. After cooling to 90° C., hot water was added and then the mixture was allowed to cool to RT. The mixture was acidified with concentrated HCl to pH=3-4. The precipitate was collected by filtration, rinsed with water (2×10 mL) and dried in vacuo to afford the product pyrido[3,2-d]pyrimidine-2,4-diol 235.
