To a solution of methyl 4-(5-methoxy-7-phenylimidazo[1,2-a]pyridin-3-yl)thiophene-2-carboxylate (94 mg, 0.26 mmol) in methanol (10 ml) and water (0.5 ml) was added a 5M aqueous solution of sodium hydroxide (0.50 ml, 2.5 mmol). The reaction mixture was heated to 70° C. After 1 h the reaction mixture was cooled and quenched with a 1N solution of hydrochloric acid (2.5 ml, 2.5 mmol), and then partially concentrated to precipitate yellow solids. The solids were washed with water and dried under vacuum to give 4-(5-methoxy-7-phenylimidazo[1,2-a]pyridin-3-yl)thiophene-2-carboxylic acid. LRMS (ESI) calculated for C19H15N2O3S [M+H]+, 351.1 found 351.0.
