1-(3,3-Dimethoxy-propyl)-5-(2-chloro-5-fluoro-pyridin-3-yl)-1H-pyrimidine-2,4-dione (Prep72, 100 mg, 0.29 mmol) was dissolved in THF (3 ml) and 2N HClaq (0.5 ml) was added. The solution was stirred at room temperature for 1 hour. THF was evaporated under vacuum and the aqueous residue was lyophilized to give 90 mg of a yellow solid that was used in the next step without further purification (93% yield).
