A stirred suspension of methyl 4-(6-amino-2-methylpyridin-3-yl)benzoate (D7, 1.0 g, 4.1 mmole) in dichloromethane (80 ml) was treated with pyridine (0.66 ml, 8.2 mmole), followed by 4-chlorobutyryl chloride (0.47 ml, 4.3 mmole). The resulting solution was stirred at 25° C. for 2 h, then treated with 5% Na2CO3 solution and after 30 minutes, the organic layer was separated, dried (Na2SO4) and concentrated in vacuo to leave a pale yellow solid. This was recrystallised from ethyl acetate/60-80 petrol to afford the title compound as a beige solid (1.13 g, 80%).
