Reaction Steps
A PNA or PNA derivative having a single free NH2 such as H2N-GCAT-COOH is dissolved in tetrahydrofuran:water to provide a solution that is 0.1 M in PNA and to this is added fluorescein isothiocyanate to give a solution which is 0.1-1.0 M in fluorescein isothiocyanate. The solution is. stirred for 0.1-2 hours, then evaporated to a solid and purified by preparative HPLC.
