A solution of {(R)-1-[(S)-2-Benzyl-4-(3-cyclopentyloxy-4-methoxy-phenyl)-piperazine-1-carbonyl]-2-hydroxy-2-methyl-propyl}-carbamic acid tert-butyl ester intermediate in CH2Cl2 (1 mL) at 0-5° C. was treated with trifluoroacetic acid and stirred at room temperature for 8 hr. After this time the reaction mixture was evaporated and the residue partitioned between EtOAc (5 ml) and a saturated aqueous solution of NaHCO3 (5 mL). The organic extract was further washed with brine then dried over MgSO4, filtered, and evaporated to afford the title compound as an oil (69 mg, 83%). LC/MS (Method B) 2.97 min, [M+1]+ 482.
