Procedure
In a 500 mL round-bottomed flask, 2-(6-tert-butyl-8-fluoro-1-oxophthalazin-2(1H)-yl)-6-(5-(5-(dimethylcarbamoyl)pyridin-2-ylamino)-1-methyl-6-oxo-1,6-dihydropyridazin-3-yl)benzyl acetate (1.142 g, 1.79 mmol) was combined with dioxane (25 ml) and 1M LiOH (10 ml) to give a light yellow suspension. The reaction mixture was stirred at 25° C. for 17 h. The crude reaction mixture was concentrated in vacuo. The residue was partitioned between DCM and water. An emulsion was obtained and the aqueous was extracted exhaustively with DCM. The organic layers were dried over Na2SO4 and concentrated in vacuo. The crude material was purified by flash chromatography (silica gel, 120 g, 0% to 4% MeOH in DCM) to afford 0.471 g (44%) of the title compound as a white solid. (M+H)+=598 m/e. 1H NMR (300 MHz, CHLOROFORM-d) δ: 8.68 (s, 1H), 8.46 (d, J=2.6 Hz, 2H), 8.29 (d, J=2.6 Hz, 1H), 7.79 (dd, J=8.3, 2.3 Hz, 1H), 7.64-7.69 (m, 1H), 7.53-7.63 (m, 3H), 7.50 (t, J=2.1 Hz, 1H), 7.47 (d, J=1.5 Hz, 1H), 7.00 (d, J=8.7 Hz, 1H), 4.43 (s, 2H), 3.92 (s, 3H), 3.11 (s, 6H), 1.43 (s, 9H).
