A mixture of 4-[(2R)-2-hydroxy-2-(4-methyl-1-oxo-1,3-dihydro-2-benzofuran-5-yl)ethyl]piperazin-2-one (26 mg, 0.089 mmol), 3-Chloro-8-methylisoquinoline-7-carbonitrile [I-12] (18 mg, 0.089 mmol), Pd2(dba)3 (16 mg, 0.017 mmol), Xantphos (21 mg, 0.036 mmol), and Cs2CO3 (40 mg, 0.12 mmol) in dioxane (4 mL) was sealed in a microwave tube and purged three times with nitrogen. It was heated to 100° C. for 10 minutes in a microwave reactor. LC showed formation of the desired product, which was purified by silica gel flash chromatography. LC-MS (IE, m/z): 457 [M+1]+.
