A mixture of 4-[(2R)-2-hydroxy-2-(4-methyl-1-oxo-1,3-dihydro-2-benzofuran-5-yl)ethyl]piperazin-2-one (50 mg, 0.17 mmol), 6-Bromo-1-benzothiophene-2-carbonitrile [I-11] (41 mg, 0.17 mmol), Pd2(dba)3 (32 mg, 0.034 mmol), Xantphos (40 mg, 0.069 mmol), and Cs2CO3 (84 mg, 0.26 mmol) in dioxane (1 mL) was sealed in a microwave tube and purged three times with nitrogen. It was heated to 120° C. for 20 minutes in a microwave reactor. LC showed formation of the desired product, which was purified by silica gel flash chromatography. LC-MS (IE, m/z): 448 [M+1]+.
