Procedure
To a clear solution of 6-[3-formyl-4-(4,4,5,5-tetramethyl-[1,3,2]dioxaborolan-2-yl)-phenoxy]-2-[(2-methoxy-ethyl)-methyl-amino]-nicotinonitrile (7) (15 g, 343 mmol) in MeOH (anhydrous, 300 mL) was slowly added NaBH4 (7.83 g, 206 mmol). The reaction was stirred at room temperature 4 hours, before the addition of HCl solution (1 M, 200 mL). The stirring was kept at room temperature overnight. Then the solution was slowly evaporated in vacuo. Purification was accomplished by reverse phase Biotage with 10%-100% MeOH/H2O gradient to afford 3 g (26% yield) of the title compound. 1H NMR (400 MHz, CHLOROFORM-d) δ ppm 7.74 (d, J=8.6 Hz, 1 H), 7.70 (d, J=8.2 Hz, 1 H), 7.13-7.09 (m, 2 H), 6.23 (d, J=8.2 Hz, 1 H), 5.08 (s, 2 H), 4.78 (s, 1 H), 3.56 (t, J=5.7 Hz, 2 H), 3.36 (t, J=5.7 Hz, 2 H), 3.27 (s, 3 H), 3.23 (s, 3 H); ES-MS: m/z 340 (M+H)+; HPLC: 99.38% (220 nm), 98.71% (MaxPlot).
